Symplex™ is Symphogen’s proprietary technology for monoclonal antibody discovering. The Symplex™ discovery process relies on isolating comprehensive repertoires of antibody variable gene pairs (known as cognate pairs) from single B lymphocytes, which are responsible for making antibodies, from naturally immune or immunized donors currently covering human, mouse or chicken. A Symplex™ repertoire of variable gene pairs thus represents a snapshot of the donor antibody responses at the time of donation. To identify single antibodies with a desired specificity or function each clone within the repertoire is expressed in microliter volumes and screened using appropriate automated assays.
The true power of the Symplex™ lies in the combination of having cognate pairs of antibody variable genes and unlimited screening of variable gene repertoires. This allow for exhaustive screening of any donor material leading to large panels of stable and high-affinity monoclonal antibodies. In case of human donors, such antibodies are further predicted to be safe for therapeutic use as they have already been counter selected for auto-reactivity by the donor immune system.
Antibody leads are identified from compositions of original antibody gene pairs (known as cognate pairs) by high throughput screening for antigen specificity.
The Symplex™ technology provides potent and safe drug leads, because the antibodies have been generated in humans and mimic the natural immune response. All processes leading to the isolation of specific antibodies have been automated, thus, the Symplex™ technology is a fast method to generate naturally high affinity drug leads.
Another benefit of this technology is its ability to identify not only polyclonal, but also monoclonal antibody drug leads, depending on the complexity of the target.
